As anyone who plays this genre we all know that there are the basics that need to be covered and then the extras we would like to have. Personally I always cover the basics, one tank (warrior/barb), 1 healer obviously, 1 mage, and one rogue, and then generally one fun class which would cover a lock or bard or druid or something hybridy that gives me some options and flexibility.

But a 4 person party precludes at least for me the fun class. Basics have to be covered. I just think a 5th party member gives players the ability to refine a party instead of just covering the basics.

Another reason is the companions strangely enough. I almost always play a mage/sorcerer for my created character. But I ran into the problem that I like Gale and hate Laz. And as such I find that my choice was pretty much felt I had to choose playing a warrior. I know I really didn't have to but to maintain the balance of my party and not have to deal with Laz I needed a tank. A 5th party member gives me the option of playing something I want as opposed to something I think I need for functionality. Though in fairness and to give credit, this is one of the first games where I actually don't mind the companions instead of trying to find a work around to make 4 custom characters.

I wouldn't mind if there was room for 6 in the party, but 5 would still be an improvement. As you say, a bigger party brings more variation. I doubt though that we will get a 5th party member as the whole game would thus have to be changed to fit that.

My basics are fighter/wizard/healer, leaving out the rogue since I don't care much for that class, and for my main character I either choose a fighter (toughest class) or a dragon blood sorcerer (high charisma, good for negotiations AND nice enough to have in a fight).

I also like to play druid, but I don't feel there is room for a druid in the current composition. One fighter (Lae'zel) + one mage (Gale) is not enough for my combat needs.

Even for a custom party I'd like a fifth slot because one slot is always occupied by a Druid or Cleric because of the Guidance cantrip...

You don't need to play the holy Trinity in D&D. Try playing how you want to during the beta. I'm doing a run now without the githyanki. And for now I have healing words in case someone goes down. In DnD you should be able to make a party of four of any classes work.

I'd like at least 6 custom party members, personally. It would be nice to have that and without mods. +The game is in early access so its the perfect time to test it.

Agreed.

I've always looked at my D&D parties as:
Tank
Melee damage-dealer
Missile damage-dealer
AoE damage-dealer
Crowd-controller
Buffer-healer

Thus, party of six is best.
