Out of pinciple I normally eradicate Sacred Stone.

So I attack the 2 singing enchantresses in the middle of the village. First of all I can attack them with poison clouds and fireballs as I want - no reaction. And second something very weird (in 3 attempts now): 1 of them does not take any further damage after her HP are halved. Ice shard, Fire, Lightning Bolt - all hits perfect but nothing does any further damage.

Anyone any idea?

Ooops. Forget the post. Found the reason. The reaction of friendly NPCs starts a dialogue even with a non main character (in this case it was the new ranger companion) and as long as this lasts the dialogue partner takes no damage. I did not notice as I switched to the next of my characters fast.

Did she cast Invulnerability on herself?

One thing that I have noticed is that even if you manage to kill the patrolling guards or the teacher/students in one shot from sneaking, it turns the rest of the town hostile. It's still possible to keep ambushing them without retaliation or reputation loss as long as you're able to kill them in one hit without being detected.

No matter, I wanted to eradicate this specific village in any case. No innocents.

Still I think the inability to kill why in a dialogue is kind of a bug. And I am not sure if not the same is true for NPCs directly addressing a companion - never happens outside hurts with weapons or spells.
