It probably depends on how famous the story is. Neon Genesis: Evangellion is based around Hebrew religion. That series is incredably popular in Japan and America and undoubtedly other countries even though the percentage of people in those countries that follow Hebrew religion is probably a lot less than other religions. A lot of the references are lost, be because of the incredable story telling, it doesn't matter. Knowledge of the Hebrew religion (I'm really sorry, but I don't know what the common name for it is.) is not essential, although it does expand the story for those who do know it.

After watching The Matrix: Revolutions, I looked up the little Indian girl's name, Sati, on the 'Net to see what significance it had. It's from Indian religion, which I know next to nothing about, and after reading the description, it fits very well into the movies. They're extra puzzles for those who want them.
