Like most funny things, the humour rather disappears if it has to be explained. But in English a doe is a female deer.
And 'Doe' sounds the same as the famous 'stupid' sound, usually spelled "D'oh". I think that it was made famous by Homer Simpson, although it had been around longer than that.
It's quite a funny caption on its own, but it was extra funny coming right after the caption in the post before, where the guy didn't see a "deer crossing" sign. <img src="/ubbthreads/images/graemlins/ROFL.gif" alt="" />
