Did you just get sent to Yarun by Sir Dante? If so, when Yarun called a couple of assassins to attack you and vanished, he was standing beside a cupboard which had previously been locked....

Have you talked to Commander Ralph in Rivertown yet (building south of the castle), and gotten the quest to investigate the old duke's murder? This quest is described in detail towards the bottom of this page: Verdistis.

There's no rush to get to the Assassins' Guild, just play the game thoroughly and you'll get a related quest sooner or later.
