I lose 2 reputation when I join the Thieves Guild. Do I need to join this guild or is it optional? If I join the Thieves Guild will I be able to join any other guilds as well, or just the one?

You can solve some quests when you join the thieves guild (but you must not do it) and you can also join other guilds after you become a member of the thieves guild.

You don't need to join the guild to follow the main story, but there are several minor quests you can't solve without becoming a member.
IIRC there are only two guilds you can join - Warriors' and Thieves' - but you can be a member of both at the same time.
You do not need to join either in order to complete the game, however.

With the latest patch, Finnigan's magic lockpicks are unbreakable, and give a +20 Agility bonus. If you have given them back, this could be considered cheating, but FYI; How to get Finnigan's lockpicks back

Why, why, I gave them to him, darn, I missed something didn't I? <img src="/ubbthreads/images/graemlins/silly.gif" alt="" /> They're just very nice to keep, they up your agility stats some plus they're unbreakable (Someone said don't put them down though). No more buying lockpicks while you have them is my understanding. <img src="/ubbthreads/images/graemlins/biggrin.gif" alt="" />

Actually, I think you HAVE to join the thieves' guild. You should anyway, but aren't they the only way to rescue Zandalor after you break out of Iona's dungeon? <img src="/ubbthreads/images/graemlins/think.gif" alt="" />

The sewer entrance is out in plain sight, and you can simply explore the sewers to find what you need there.
The Thieves' Guild is not essential at any point.

guild: I find it nice to enter as many guilds as possible, although if you want to get the experience or the house, the merchant's guild is unavoidable, imho. Thieve's guild's nice, and it gives you that little lockpicking contest too, which I haven's solved yet personally, so I don't know if it's really worth it or not. lockick: well, I've got 56 lockpicks in my inventory right now, and I can't find a way to get rid of them really, they're quite heavy. 86 super healing potions count up to aprxm 500 in weight too, and I'd like to find a way to separate them in half at least, cause all those plus the lockpicks plus the super mana potions (another 70), I can't carry anything around really. Thank god charms don't weigh anything <img src="/ubbthreads/images/graemlins/silly.gif" alt="" />

Lockpicks: 1. Sell them or 2. Open your inventory, drag the lockpicks out of it and before you let go of the mouse, press CTRL. A pop up window appears, the one you know from trading Kiya
Raze explains it like this Drop the lockpicks on the ground, then drag them into your inventory and hold the control key down before dropping. This will bring up the split stack pane, which will let you specify how much lockpicks to drop into your inventory.

have you got finnigans lockpicks? Don't give them back to im because they give ya +20 agility and have unlimited use. And i suggest you should use restoration potions, they give you -minor 50% mana/vit back and -normal 100% mava/vit back. Super res. potion is useless.
If you are in a sticky situation then the best tactic is to get out of it.

Winning the lockpicking contest allows you to loot the Thieves Guild treasury, so yes, it's worth doing <img src="/ubbthreads/images/graemlins/smile.gif" alt="" />

What was this about buying a house from the Merchant's Guild? I have the option now, but wasn't sure if I wanted to spend that kind of money. I think I may though, if I can buy one to use as a home base and can decorate it instead of one way back in Aleroth.

I suggest to buy the house, you get some exp. You can also rent is much cheaper, but after 30 days you won't be able to use the bed anymore and I wasn't able to renew the rent or buy the house after the rent expired. next time i will certainly buy. Don't worry about money you will have more than enough
