As Daft mentions you skip hours of gameplay and other things like skill books.
Also the followers of the battle tower, the ones you hire in sentinel islands, give quests involving killing the leaders of those fortresses. Beside quest rewards they upgrade their area.
Nonetheless I do not remember any quest that is part of the main quest.
