After talking to Desdemona, an exclamation mark shows up above Madoras' head. When I click to talk to her, nothing happens. Additionally, now I can't talk/interact with any NPC.

I had the exact same problem, although wandering out to the town gate and back again seemed to cure the NPC side of it. I still can't talk to Madora though...but I haven't tried heading to the lighthouse yet which I'm assuming will clear it.

EDIT: ah well, so much for that assumption! I've completed the Desdemona quest and Madora is still mute. Other NPC interactions are fine though.

Went to lighthouse already and entered lower floor, both ghosts are present but there is no dialogue option. I think I'm fubared!
I'll try exiting/entering city. If that doesn't work I'll have to reload save just before locked door and fight naties all over again. Oh well....

I've had this first time I talked to Desdemona, after I teleported away.
I reloaded that save after talking to her, teleported again, nothing.

So yeah, something odd is definitely going on there, but if you have a save around there you can try loading it and see if it happends again?

Well, I tried exiting Cyseal and re-entering and yes I was able to interact with NPC's, but only my Male character's. The female character's were mute! I tried several city exits/enters, still bugged.

So... reload previous Black Cove early save, entered/cleared Ominous Cove, spoke with Desdemona and after she pops away the exclamation mark showed up again. Bug is definately repeatable so please take note. I guess I can't progress further in the story since I have limited/no dialogue options.

Did you try separating your female characters and exiting the city just with them, or just the female main character?

Do you still have the saves where you couldn't talk to NPCs, or with that half fixed fixed?
If so, please email supportdos@larian.com, with a description of the problem and the report.zip file generated by the D:OS support tool. The zip file will contain some system information, configuration files, and you saves (if you have a lot of saves you can delete unnecesary one from the zip file, or move them out of the '..\Documents\Larian Studios\Divinity Original Sin\Savegames' folder before creating the report)

I have multiple saves where Madora has the lovely yellow exclamation point above her head....but the AnalysisTool doesn't work for me. Every time I run it (even as administrator) it falls over.

Thanks for trying, but I don't think that's necessary. The problem with Desdemona, Madora and not being able to talk to other NPCs is known (and I think should be fixed in the next update). I don't recall seeing Fenix66d's issue before, though (male characters being able to talk to NPCs and female ones not), so thought it wouldn't hurt to send a save of that to support (don't know if that variation would have been obvious when they fixed the main issue, though it is probably irrelevant).

Just a follow-up to my original posts. I started a new playthrough and reached this quest again. As noted the problem with Desdemona and Madora was encountered again. I teleported to the Cyseal North Rift Gate, went to the market square area and tried to interact with the Potion Vendor (others too). Again Scarlett was unable to interact with any NPC in the city but this time neither could Roderick. Only Jahan was able to interact with any NPC vendor.

Read in another thread that exiting via the East Gate and then re-entering the city will fix talking issue with vendors. Tried this and it did indeed work, except for Madora which is a known issue and results in the Lost Love at the Lighthouse quest from being completed..
