You need to select both the target (you can use the character portraits on the left side of the screen) and the destination. Immediately after selecting a character by clicking on them you can get a 'Target is blocked' message, but if you move the cursor away somewhere that is clear and in their line of sight, the message should change to 'Choose teleport destination'.
