Are you using a keyboard that has alternate functions assigned to the F keys? You may need to hit an Fn key, or something, to get them to behave as function keys.
If you click on the 'Browse Skills' icon (or Browse Potions) in the top right of the UI at the bottom of the screen, it will open a menu showing all your active and passive skills, your class special move and (once you have them) actions for the teleporter pyramids.
With the mouse cursor hovering over an active skill (or potion) pressing a function key should assign it to that spell (potion) and put the corresponding F# in the bottom let corner of the icon.
After that, pressing the function key should switch to that spell (make it active) so you can then right click to use the spell (potions are used immediately when their hotkey is pressed).
