The door is actually checking to see if all of the portals in the homestead have been opened (except for the Forbidden Zone one).
You can not sell blood stones, and given their use provides experience, a full heal and resurrects any dead party members (plus used for a couple of side quests), even without the connection to opening portals in the homestead, they shouldn't be discarded.
The inert stones are not required, though if you keep them it does make it easier to keep track of how many blood/star stones you have found.
Check the homestead, make sure you stop the attack if you have not already done so, look for Zixzax and see if he is hanging around by the statue or a portal waiting for your main characters to open the next portal, or waiting at the last opened portal for you to enter, get the cutscene conversation with Astarte or the demon and leave through the portal to get back to the main section of the homestead. A map marker is added when a portal is completely open, so if there is an active portal with no map flag, that is the one holding up opening any further portals.
It is also possible to bypass the door in the Source temple.
Drop a teleporter stone in front of the door (I got it to work dropping on the left side, a few steps back), then drag and drop it from the past the door (rotate the camera to see; you might need to move the cursor around a bit to find a place where the pyramid is outlined in white and you can drop it).
