Right now, for Astarion, I'm thinking:

3 Thief
6 Fighter (two weapon fighting)
3 Gloom Stalker

Of course, the story may take me in a different direction, who knows? But all else being equal, this is what I'm thinking for him in my first playthrough. I might even do his story first; I find it interesting.

Do you have any ideas of what Astarion will look like in your universe?

You can get two weapon fighting from ranger level two, so I'd stick with either fighter or ranger. If you're set on mixing these classes though, I'd go 2 Rogue / 5 Gloom / 5 fighter of choice; that way you still get the extra attacks from the warrior ethos, and sacrifice very little from one thief level.

To be fair, my first char will be a Gloom 5 / Rogue 7 so I'll not go the same route with Astarion.
But. Next on my list is Shadowheart and I want to play Shadowheart as a rogue multi, too - goes great with her Cleric subclass and her secret Sharran operative character. So now I have two roguey playthroughs in a row and will probably postpone Astarion for a bit. But, If I wasn't doing my own version of this, I'd say:

Assassin 7, Gloom 5.

+ Invisible in the dark to all who rely on darkvision [pretty much everything except humans or casters with True Sight] - This means you don't have to spend a bonus action to hide. Helps more for archery, but you can escape into the shadows without having to hide.

+ 3D6 Sneak Attack
+ Two attacks per round [only one sneak attack per round] + Offhand; so even if you miss once, you're going to hit once per round and get your Sneak off.
+ Assassin Crit Guarantee
+ Ranger Casting; Hunter's Mark, Jump, Speak with animals - all good stuffs.
+ All the best stuff from both classes

Edit: And, as Astarion, you can speak with animals, then eat them.

I'm still thinking Level 1 rogue.

I will make rogue 2/fighter 2/ranger 2/paladin 2/cleric 2/warlock 2 Astarion build.

Without the +1 he’s no more dead than when you meet him after all.

I think he's still on AD&D Vampires, which require +1 weapons to hit.

In my first playthrough he might just stay rogue, but when i do a dark urge who submits to temptation i might multi class him to cleric of bhaal.

Ouch... are you all already making plans for your killer Dark Urge Tav? Nope, I am not killing any party members. 😄

As for the multiclassing:
My Astarion is usually an Arcane Trickster. I'm new to multiclassing in 5E, but Gloomstalker sounds very interesting. I've got to familiarize myself more with 5E multiclassing in general, but would it work as well with the Arcane Trickster instead of the Thief?

Rogue 1
Fighter 5 (Battlemaster) (Dual wielder)

Yes, fighter 5 for the dual wield, action surge and extra attack. The extra action from thief 3 gives an extra attack per round at the moment in EA so it is hard to see how assassin (as awesome as it sounds) could beat this. But as rodeolifant said, this is only if I don't play the rogue role with my main and rogues are so much fun in EA and the gloomstalker / thief or shadow monk / thief are pretty tempting.
And as tempting as it always is to kill Astarion, the first playthrough for me is always the completionist do everyone's side story playthrough.

And unless i forgot something, it seems like you got to aproximately same Damage as you would have with Rogue on lvl 12 ...

Therefore there are two questions that radicaly affect this build imho:
- Will it be possible to deliver Sneak Attack with Off Hand Attack?
- How often will you miss?

Personaly i lean more towards builds where you try to make the strike count, rather than get more atempts to strike.

This This will net you two attacks per round [main hand] + 1 attack off hand [bonus action] + plus action surge.

Yes, but you only get one sneak attack per round.

Astarion will be dead, just like in all of my EA playthroughs, I just can't stand him so he dies the moment I see him.

I honestly don't think, that I will bother with Astarion too much, since he is my least favorite companion ( well, apart from Minthara, who probably won't be in my party at all). I'll keep him around to get his story, if he doesn't leave, because I'm too good, but I won't invest too much in him.
If I would multiclass him, then probably with gloomstalker.

Can you sneak attack multiple times in a round?
Sneak attack can only be used once per turn, but it is not limited to your turn. That means you can use sneak attack once on your turn, but subsequent attacks on other turns could also include sneak attack damage. This could include using your reaction for an attack of opportunity
