Defying Mystra does not seem to have any consequences. Just like all the tadpoles in your brain.
Spoilers for origin Gale playthrough:
If you show up to her with the crown and say you're going to challenge her, you get an anime battle staredown after which she just.. deletes you and it ends there.
...I can't say that sounds much better to me. I mean, I haven't done any origin character playthroughs so maybe that makes sense for Gale? I dunno, how did you feel about it?
